Public bus departures are mostly from the bus station on the western side of town at the junction of Minzu Lu and Chingdol Zhong Lu. It's advisable to buy tickets a day in advance - there's a foreigners' ticket office and staff are generally helpful. Pilgrim buses and minibuses leave from various points but note the possible restrictions on your use of these. Further details are given in the accounts of the destinations and in "Travel details". The situation in Tibet with regards to transport and regulations is in a constant state of flux. It's essential to talk to other travellers to try to get an up-to-date picture of things.
